    Friedel oscillations of one-dimensional correlated fermions from perturbation theory and density functional theory

    We study the asymptotic decay of the Friedel density oscillations induced by an open boundary in a one-dimensional chain of lattice fermions with a short-range two-particle interaction. From Tomonaga-Luttinger liquid theory it is known that the decay follows a power law, with an interaction dependent exponent, which, for repulsive interactions, is larger than the noninteracting value −1-1. We first investigate if this behavior can be captured by many-body perturbation theory for either the Green function or the self-energy in lowest order in the two-particle interaction. The analytic results of the former show a logarithmic divergence indicative of the power law. One might hope that the resummation of higher order terms inherent to the Dyson equation then leads to a power law in the perturbation theory for the self-energy. However, the numerical results do not support this. Next we use density functional theory within the local-density approximation and an exchange-correlation functional derived from the exact Bethe ansatz solution of the translational invariant model. While the numerical results are consistent with power-law scaling if systems of 10410^4 or more lattice sites are considered, the extracted exponent is very close to the noninteracting value even for sizeable interactions.Comment: 11 pages, 5 figure

    DatenmanagementplĂ€ne werden zunehmend als wichtiges Element der guten wissenschaftlichen Praxis verstanden und bei der Beantragung von Fördermitteln eingefordert. Die Workshopreihe „DatenmanagementplĂ€ne in der Forschung – von Grundlagen zu Grundfragen“ hatte zum Ziel, die unterschiedlichen Perspektiven auf DatenmanagementplĂ€ne zu beleuchten und einen Austausch zwischen allen am Thema Beteiligten zu ermöglichen. Hierzu wurde im ersten Workshop zunĂ€chst ein gemeinsames VerstĂ€ndnis fĂŒr Definition und Inhalte eines Datenmanagementplans geschaffen. In den zwei folgenden Veranstaltungen wurden die Perspektiven der Forschungsförderer und der Forschenden diverser Fachbereiche beleuchtet. Die abschließende vierte Veranstaltung stellte verschiedene Tools zur Erstellung von DatenmanagementplĂ€nen vor. Der vorliegende Artikel informiert ĂŒber Inhalte und Ergebnisse der vier Workshops und gibt einen Ausblick auf zukĂŒnftige Entwicklungen

    A sperm-activating peptide controls a cGMP-signaling pathway in starfish sperm☆

    AbstractPeptides released from eggs of marine invertebrates play a central role in fertilization. About 80 different peptides from various phyla have been isolated, however, with one exception, their respective receptors on the sperm surface have not been unequivocally identified and the pertinent signaling pathways remain ill defined. Using rapid mixing techniques and novel membrane-permeable caged compounds of cyclic nucleotides, we show that the sperm-activating peptide asterosap evokes a fast and transient increase of the cGMP concentration in sperm of the starfish Asterias amurensis, followed by a transient cGMP-stimulated increase in the Ca2+ concentration. In contrast, cAMP levels did not change significantly and the Ca2+ response evoked by photolysis of caged cAMP was significantly smaller than that using caged cGMP. By cloning of cDNA and chemical crosslinking, we identified a receptor-type guanylyl cyclase in the sperm flagellum as the asterosap-binding protein. Sperm respond exquisitely sensitive to picomolar concentrations of asterosap, suggesting that the peptide serves a chemosensory function like resact, a peptide involved in chemotaxis of sperm of the sea urchin Arbacia punctulata. A unifying principle emerges that chemosensory transduction in sperm of marine invertebrates uses cGMP as the primary messenger, although there may be variations in the detail

    Bewertung landwirtschaftlicher ProduktivitÀtspotentiale der globalen Landressource

    Ziel unserer Arbeiten ist die Klassifikation und funktionelle Bewertung von Agrarstandorten nach einheitlichen Kriterien. Solche Informationen könnten fĂŒr die ĂŒberregional vergleichende Bewertung und standortgerechte nachhaltige agrarische Nutzung von Böden hilfreich sein. Wir untersuchten mehr als 100 reprĂ€sentative Böden auf Versuchsstandorten in bedeutenden Agrarregionen der Welt. Das sind Europa, Nordchina, die PrĂ€rieregionen Nordamerikas, sowie die Steppen Westsibiriens und Kasachstans. Die Böden wurden nach der World Reference Base for Soil Resources (WRB 2006) klassifiziert und mittels MĂŒncheberger Soil Quality Rating (M-SQR) funktionell bewertet. Das indikatorbasierte M-SQR erwies sich als praktikabel und fĂŒhrte zu plausiblen Wertezahlen der BodengĂŒte. Die Bewertungskennziffern des M-SQR können je nach LandnutzungsintensitĂ€t und AnpassungsgĂŒte der Indikatoren etwa 50-80 % der ErtragsvariabilitĂ€t von Getreide erklÀren. Es wird geschlußfolgert, daß das M-SQR fĂŒr die skalenĂŒbergreifende AbschĂ€tzung von landwirtschaftlichen ProduktivitĂ€tspotentialen, also als Basiswerkzeug fĂŒr ein globales Bodenmonitoring, geeignet ist.     Vagal control of the heart decreases during increasing imminence of interoceptive threat in patients with panic disorder and agoraphobia

    Theoretically, panic disorder and agoraphobia pathology can be conceptualized as a cascade of dynamically changing defensive responses to threat cues from inside the body. Guided by this trans‑diagnostic model we tested the interaction between defensive activation and vagal control as a marker of prefrontal inhibition of subcortical defensive activation. We investigated ultra‑short‑term changes of vagally controlled high frequency heart rate variability (HRV) during a standardized threat challenge (entrapment) in n = 232 patients with panic disorder and agoraphobia, and its interaction with various indices of defensive activation. We found a strong inverse relationship between HRV and heart rate during threat, which was stronger at the beginning of exposure. Patients with a strong increase in heart rate showed a deactivation of prefrontal vagal control while patients showing less heart rate acceleration showed an increase in vagal control. Moreover, vagal control collapsed in case of imminent threat, i.e., when body symptoms increase and seem to get out of control. In these cases of defensive action patients either fled from the situation or experienced a panic attack. Active avoidance, panic attacks, and increased sympathetic arousal are associated with an inability to maintain vagal control over the heart suggesting that teaching such regulation strategies during exposure treatment might be helpful to keep prefrontal control, particularly during the transition zone from post‑encounter to circa strike defense

    Therapygenetic effects of 5-HTTLPR on cognitive-behavioral therapy in anxiety disorders: a meta-analysis

    There is a recurring debate on the role of the serotonin transporter gene linked polymorphic region (5-HTTLPR) in the moderation of response to cognitive behavioral therapy (CBT) in anxiety disorders. Results, however, are still inconclusive. We here aim to perform a meta-analysis on the role of 5-HTTLPR in the moderation of CBT outcome in anxiety disorders. We investigated both categorical (symptom reduction of at least 50%) and dimensional outcomes from baseline to post-treatment and follow-up. Original data were obtained from ten independent samples (including three unpublished samples) with a total of 2,195 patients with primary anxiety disorder. No significant effects of 5-HTTLPR genotype on categorical or dimensional outcomes at post and follow-up were detected. We conclude that current evidence does not support the hypothesis of 5-HTTLPR as a moderator of treatment outcome for CBT in anxiety disorders. Future research should address whether other factors such as long-term changes or epigenetic processes may explain further variance in these complex gene-environment interactions and molecular-genetic pathways that may confer behavioral change following psychotherapy

    Three-dimensional super-resolution microscopy of the inactive X chromosome territory reveals a collapse of its active nuclear compartment harboring distinct Xist RNA foci

    Background: A Xist RNA decorated Barr body is the structural hallmark of the compacted inactive X territory in female mammals. Using super resolution three-dimensional structured illumination microscopy (3D-SIM) and quantitative image analysis, we compared its ultrastructure with active chromosome territories (CTs) in human and mouse somatic cells, and explored the spatio-temporal process of Barr body formation at onset of inactivation in early differentiating mouse embryonic stem cells (ESCs). Results: We demonstrate that all CTs are composed of structurally linked chromatin domain clusters (CDCs). In active CTs the periphery of CDCs harbors low-density chromatin enriched with transcriptionally competent markers, called the perichromatin region (PR). The PR borders on a contiguous channel system, the interchromatin compartment (IC), which starts at nuclear pores and pervades CTs. We propose that the PR and macromolecular complexes in IC channels together form the transcriptionally permissive active nuclear compartment (ANC). The Barr body differs from active CTs by a partially collapsed ANC with CDCs coming significantly closer together, although a rudimentary IC channel system connected to nuclear pores is maintained. Distinct Xist RNA foci, closely adjacent to the nuclear matrix scaffold attachment factor-A (SAF-A) localize throughout Xi along the rudimentary ANC. In early differentiating ESCs initial Xist RNA spreading precedes Barr body formation, which occurs concurrent with the subsequent exclusion of RNA polymerase II (RNAP II). Induction of a transgenic autosomal Xist RNA in a male ESC triggers the formation of an `autosomal Barr body' with less compacted chromatin and incomplete RNAP II exclusion. Conclusions: 3D-SIM provides experimental evidence for profound differences between the functional architecture of transcriptionally active CTs and the Barr body. Basic structural features of CT organization such as CDCs and IC channels are however still recognized, arguing against a uniform compaction of the Barr body at the nucleosome level. The localization of distinct Xist RNA foci at boundaries of the rudimentary ANC may be considered as snap-shots of a dynamic interaction with silenced genes. Enrichment of SAF-A within Xi territories and its close spatial association with Xist RNA suggests their cooperative function for structural organization of Xi

    Evidence that breast cancer risk at the 2q35 locus is mediated through IGFBP5 regulation.

    GWAS have identified a breast cancer susceptibility locus on 2q35. Here we report the fine mapping of this locus using data from 101,943 subjects from 50 case-control studies. We genotype 276 SNPs using the 'iCOGS' genotyping array and impute genotypes for a further 1,284 using 1000 Genomes Project data. All but two, strongly correlated SNPs (rs4442975 G/T and rs6721996 G/A) are excluded as candidate causal variants at odds against >100:1. The best functional candidate, rs4442975, is associated with oestrogen receptor positive (ER+) disease with an odds ratio (OR) in Europeans of 0.85 (95% confidence interval=0.84-0.87; P=1.7 × 10(-43)) per t-allele. This SNP flanks a transcriptional enhancer that physically interacts with the promoter of IGFBP5 (encoding insulin-like growth factor-binding protein 5) and displays allele-specific gene expression, FOXA1 binding and chromatin looping. Evidence suggests that the g-allele confers increased breast cancer susceptibility through relative downregulation of IGFBP5, a gene with known roles in breast cell biology

    The Human Phenotype Ontology in 2017.

    Deep phenotyping has been defined as the precise and comprehensive analysis of phenotypic abnormalities in which the individual components of the phenotype are observed and described. The three components of the Human Phenotype Ontology (HPO; www.human-phenotype-ontology.org) project are the phenotype vocabulary, disease-phenotype annotations and the algorithms that operate on these. These components are being used for computational deep phenotyping and precision medicine as well as integration of clinical data into translational research. The HPO is being increasingly adopted as a standard for phenotypic abnormalities by diverse groups such as international rare disease organizations, registries, clinical labs, biomedical resources, and clinical software tools and will thereby contribute toward nascent efforts at global data exchange for identifying disease etiologies. This update article reviews the progress of the HPO project since the debut Nucleic Acids Research database article in 2014, including specific areas of expansion such as common (complex) disease, new algorithms for phenotype driven genomic discovery and diagnostics, integration of cross-species mapping efforts with the Mammalian Phenotype Ontology, an improved quality control pipeline, and the addition of patient-friendly terminology
